How were you manipulating the permissions?
I find that the stock query works fine:
GRANT SELECT, INSERT,DELETE on mydb.* to systemuser@'%' ;
GRANT SELECT, INSERT,DELETE on mydb.* to systemuser@'localhost';
although you may want to play around with allowable networks to connect from.
More typically, I just do
GRANT ALL on mydb.* to mydb@'%' ;
does the job nicely.
